Background: it has been proposed that the hepatic glycolytic blockade associated with a hemizygous PFKFB1 variant (p.Ser159Thr) causes cytosolic NADH accumulation, with a secondary increase in demand on the malate-aspartate shuttle (MAS) — the principal mechanism for transferring reducing equivalents from the cytosol to the mitochondria in the hepatocyte. In the index case (companion report […]

Introduction: Autism spectrum disorder (ASD) is a neurodevelopmental condition characterised by high aetiological and clinical heterogeneity. In recent years, the need to investigate integrative approaches that account for its possible multisystemic nature has been increasingly recognised. From a complex systems perspective, effective interventions should be necessarily multidimensional and adaptive. Case presentation: A 3-year-old male participant […]

In recent years, there has been growing recognition of the need to investigate integrative approaches that consider its potential multisystem nature. Introduction: Mothers of children with autism spectrum disorder (ASD) experience sustained functional impairment across multiple life domains, including sleep, autonomy, health, and social participation. Despite growing evidence on caregiver burden in ASD, the impact of child-directed interventions on maternal quality of life has not been systematically documented. This study explores perceived functional changes in primary […]

Introduction: Mothers of children with autism spectrum disorder (ASD) experience sustained functional impairment that simultaneously affects all areas of daily life. Available instruments for assessing this impact, designed within the clinical-psychological framework, capture internal states such as stress or anxiety but fail to measure the mother’s functional capacity to sustain her own life. Naturopathy, through […]

The Katia Dolle Method (KDM) is a hierarchical clinical decision framework developed within clinical naturopathy for multisystemic intervention in neurodevelopmental disorders, with primary application in autism spectrum disorder (ASD). Developed over more than twenty-five years of intensive clinical practice and applied to a population exceeding 1,000 users across 32 countries, the KDM integrates principles from […]

Background: Refractory obsessive-compulsive disorder (OCD) represents a major clinical challenge. We describe the case of a 34-year-old male (F.R.C.) with severe OCD (baseline Y-BOCS = 24/40), childhood onset, eleven years of partial pharmacological response and documented hypersensitivity to multiple psychotropic drugs. The baseline qEEG (T0, 02/10/2025) was consistent with PANDAS with OCD comorbidity and emotional […]

Background: PFKFB1 (6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase 1) is a bifunctional enzyme regulating hepatic glycolytic flux. No OMIM disease entity is currently associated with PFKFB1 variants, and the clinical literature is virtually nonexistent. We describe the first prospective interventional case study in a participant carrying a hemizygous PFKFB1 variant (p.Ser159Thr, ChrX:54984780A>T). Case: A 14-year-old male with myoclonic epilepsy, a […]

This report presents a longitudinal single-case study of a child with autism spectrum disorder (ASD) undergoing a multimodal naturopathic intervention based on the Katia Dolle Method® (KDM). The intervention was designed within a complex systems framework, targeting interconnected biological factors including intestinal dysbiosis, food hypersensitivity, micronutrient imbalances, and heavy metal burden. The case involves a […]

This retrospective cross-sectional study evaluates the effectiveness of the Katia Dolle Method® in reducing the severity of Autism Spectrum Disorder (ASD) in 183 children. The analysis was based on pre- and post-intervention ATEC scores collected between 2017 and 2024. Results show a highly significant reduction in ATEC scores (mean reduction: 39.20 points; p < 0.001), […]
